The substance being analyzed is usually a protein or antigen. The ELISA plate is generally coated with an antibody that is specific to the substance of interest. The antibody will then bind the substance that is present in the sample and any non-specific substances can be washed away. Most ELISAs use a HRP-based enzymatic reaction to produce a color change. The intensity can be quantified using a spectrophotometer and reflects the amount of the substance of interest that is present in the sample. The OD values of the samples can then be compared to the OD values of a set of standards with known concentrations to determine the concentration of the substance of interest in the sample.
Monoclonal antibodies can be raised against almost any substance, and they will then bind to that specific substance to aid in the detection or purification. This is important for the fields of medicine, biochemistry and molecular biology. Compared to monoclonal antibodies which bind to specific epitope and can be too specific, polyclonal antibodies bind to multiple epitopes to offer greater sensitivity for detecting proteins that show minor variations in individual epitopes.
Antibodies against the immunogen can then be purified from the serum of the animal. The purified fluid will contain a mixture of polyclonal antibodies that will bind to various epitopes throughout the immunogenic sequence. Polyclonal antibodies are typically less expensive to produce, and they are generally more suitable for applications in which binding to multiple epitopes will produce more favorable results, such as IP/ChIP.
While this can be a laborious process, the result is an isolated, specific protein. The process often involves a combination of many methods that exploit differences between various proteins in binding affinity, physico-chemical properties, size and biological activity. Secondary antibodies are often conjugated to substances that will enable the visualization of the primary antibody, such as HRP or fluorophores. The Fab domain binds to the Fc domain of the primary antibody and because the Fc domain is consistent within the same animal class, a single type of primary antibody can bind many types of primary antibodies as long as the host species is the same.
They are distinguished from proteins based on size, and a chain of 50 or fewer amino acids is generally considered to be a peptide. Peptides are commonly used in molecular biology for various applications. They can be used as immunogens to produce antibodies against specific regions of proteins, and this eliminates the need to purify native proteins.
