 Specificity    This assay has high sensitivity and excellent specificity for detection of human ATG12. No significant cross-reactivity or interference between human ATG12 and analogues was observed.
 Samples    For the quantitative determination of human ubiquitin-like protein ATG12 (ATG12) concentrations in serum, plasma, tissue homogenates, cell lysates.
 Detection Range    25 pg/ml -1600 pg/ml.
 Sensitivity    The minimum detectable dose of human ATG12 is typically less than 6.25 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
 Intra-assay Precision    Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
 Inter-assay Precision    Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
 Preparation and Storage    Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
 Other Notes for ATG12 elisa kit    Small volumes of ATG12 elisa kit may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice.

Product Description specifically for ATG12 elisa kit

   Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ATG12 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ATG12 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ATG12 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ATG12 bound in the initial step. The color development is stopped and the intensity of the color is measured.

 NCBI Summary for ATG12    Autophagy is a process of bulk protein degradation in which cytoplasmic components, including organelles, are enclosed in double-membrane structures called autophagosomes and delivered to lysosomes or vacuoles for degradation. ATG12 is the human homolog of a yeast protein involved in autophagy (Mizushima et al., 1998 [PubMed 9852036]).[supplied by OMIM, Mar 2008]

Conjugation with ATG5 through a ubiquitin-like conjugating system involving also ATG7 as an E1-like activating enzyme and ATG10 as an E2-like conjugating enzyme, is essential for its function. The ATG12-ATG5 conjugate acts as an E3-like enzyme which is required for lipidation of ATG8 family proteins and their association to the vesicle membranes.
