MBS2020230 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Alpha-2-Macroglobulin (a2M) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing a2M. The ELISA analytical biochemical technique of the MBS2020230 kit is based on a2M antibody-a2M antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect a2M antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, a2M. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: The kit is a competitive inhibition enzyme immunoassay technique for the in vitro quantitative measurement of a2M in mouse serum, plasma, urine and other biological fluids.

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific to a2M has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled a2M and unlabeled a2M (Standards or samples) with the pre-coated antibody specific to a2M. After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of a2M in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of a2M in the sample.

Alpha-2-macroglobulin is a protease inhibitor and cytokine transporter. It inhibits many proteases, including trypsin, thrombin and collagenase. A2M is implicated in Alzheimer disease (AD) due to its ability to mediate the clearance and degradation of A-beta, the major component of beta-amyloid deposits. [provided by RefSeq, Jul 2008]

A2M: Is able to inhibit all four classes of proteinases by a unique 'trapping' mechanism. This protein has a peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molecular weight substrates (activity against high molecular weight substrates is greatly reduced). Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase. Belongs to the protease inhibitor I39 (alpha-2- macroglobulin) family.
