The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ACER2. The ELISA analytical biochemical technique of the MBS7209609 kit is based on ACER2 antibody-ACER2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ACER2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ACER2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This ACER2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse ACER2. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: ACER2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ACER2 antibody and an ACER2-HRP conjugate. The assay sample and buffer are incubated together with ACER2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACER2 concentration since ACER2 from samples and ACER2-HRP conjugate compete for the anti-ACER2 antibody binding site. Since the number of sites is limited, as more sites are occupied by ACER2 from the sample, fewer sites are left to bind ACER2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACER2 concentration in each sample is interpolated from this standard curve.

The sphingolipid metabolite sphingosine-1-phosphate promotes cell proliferation and survival, whereas its precursor, sphingosine, has the opposite effect. The ceramidase ACER2 hydrolyzes very long chain ceramides to generate sphingosine (Xu et al., 2006 [PubMed 16940153]).[supplied by OMIM, Jul 2010]
