The ELISA analytical biochemical technique of the MBS8244709 kit is based on ADIPOQ antibody-ADIPOQ antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ADIPOQ antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ADIPOQ. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the Assay: MBS Rat Adiponectin ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat Adiponectin in Cell Culture Supernatants, Serum, Plasma, Tissue Homogenates, Urine. This assay employs an antibody specific for Rat Adiponectin coated on a 96-well plate. Standards and samples are pipetted into the wells and Adiponectin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat Adiponectin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Adiponectin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: Adiponectin (ADPN) is a hormone secreted by adipocytes that regulates energy homeostasis and glucose and lipid metabolism. Adiponectin is a new member of the family of soluble defense collagens, in hematopoiesis and immune responses. It is an important negative regulator in hematopoiesis and immune systems and raise the possibility that it may be involved in ending inflammatory responses through its inhibitory functions. Adiponectin is mapped to 3q27 and can protect the organism from systemic inflammation by promoting the clearance of early apoptotic cells by macrophages through a receptor-dependent pathway involving calreticulin.
