The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ADORA2a. The ELISA analytical biochemical technique of the MBS2022744 kit is based on ADORA2a antibody-ADORA2a antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ADORA2a antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ADORA2a. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Adenosine A2a Receptor (ADORA2a). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Adenosine A2a Receptor (ADORA2a). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Adenosine A2a Receptor (ADORA2a), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Adenosine A2a Receptor (ADORA2a) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a member of the guanine nucleotide-binding protein (G protein)-coupled receptor (GPCR) superfamily, which is subdivided into classes and subtypes. The receptors are seven-pass transmembrane proteins that respond to extracellular cues and activate intracellular signal transduction pathways. This protein, an adenosine receptor of A2A subtype, uses adenosine as the preferred endogenous agonist and preferentially interacts with the G(s) and G(olf) family of G proteins to increase intracellular cAMP levels. It plays an important role in many biological functions, such as cardiac rhythm and circulation, cerebral and renal blood flow, immune function, pain regulation, and sleep. It has been implicated in pathophysiological conditions such as inflammatory diseases and neurodegenerative disorders. Alternative splicing results in multiple transcript variants. A read-through transcript composed of the upstream SPECC1L (sperm antigen with calponin homology and coiled-coil domains 1-like) and ADORA2A (adenosine A2a receptor) gene sequence has been identified, but it is thought to be non-coding.
