This AFP ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse AFP. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of AFP elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS730245 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Alpha-fetoprotein (AFP) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing AFP. The ELISA analytical biochemical technique of the MBS730245 kit is based on AFP antibody-AFP antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect AFP antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, AFP. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%). Unlike fetoprotein AFP human, rodent alpha fetoprotein AFP binds and transports estrogen across maternal placentas and effects sexual development of fetal mouse. Thus, quantitation of this key plasma protein in research settings is of great interest both in reproductive and cancer antigen biology. We are able to provide a highly sensitive competitive format fetoprotein AFP ELISA perfect for your assay requirements. Both our mouse specific and human alpha fetoprotein ELISA kits are shipped complete after your order. All necessary reagents for successful results from serum, cell culture media or cell lysates from this product are included - AFP monoclonal antibody clone, secondary polyclonal antibody for detection with ALEXA 174 fluor or AMP. All our kits reagents are molecular biology grade, making comparative studies with FISH probes or q PCR possible. This ELISA kit, like all of our products, either standard or custom antibodies, are fully satisfaction guaranteed. View online today.

Principle of the Assay: AFP ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AFP antibody and an AFP-HRP conjugate. The assay sample and buffer are incubated together with AFP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AFP concentration since AFP from samples and AFP-HRP conjugate compete for the anti-AFP antibody binding site. Since the number of sites is limited, as more sites are occupied by AFP from the sample, fewer sites are left to bind AFP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AFP concentration in each sample is interpolated from this standard curve.

NCBI Summary for AFP
This gene encodes alpha-fetoprotein, a major plasma protein produced by the yolk sac and the liver during fetal life. Alpha-fetoprotein expression in adults is often associated with hepatoma or teratoma. However, hereditary persistance of alpha-fetoprotein may also be found in individuals with no obvious pathology. The protein is thought to be the fetal counterpart of serum albumin, and the alpha-fetoprotein and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. Alpha-fetoprotein is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. The level of alpha-fetoprotein in amniotic fluid is used to measure renal loss of protein to screen for spina bifida and anencephaly. [provided by RefSeq, Jul 2008]
UniProt Comments for AFP
AFP: Binds copper, nickel, and fatty acids as well as, and bilirubin less well than, serum albumin. Only a small percentage (less than 2%) of the human AFP shows estrogen-binding properties. Dimeric and trimeric forms have been found in addition to the monomeric form. Plasma. Synthesized by the fetal liver and yolk sac. Belongs to the ALB/AFP/VDB family.
