Species Reactivity
This assay has high sensitivity and excellent specificity for detection of AGRIN. No significant cross-reactivity or interference between AGRIN and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between AGRIN and all the analogues, therefore, cross reaction may still exist in some cases.
Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate
Assay Type
Competitive or Sandwich
Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of Agrin elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS721753 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Agrin, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing Agrin. The ELISA analytical biochemical technique of the MBS721753 kit is based on Agrin antibody-Agrin antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Agrin antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, Agrin. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This AGRIN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human AGRIN. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: AGRIN ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AGRIN antibody and an AGRIN-HRP conjugate. The assay sample and buffer are incubated together with AGRIN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AGRIN concentration since AGRIN from samples and AGRIN-HRP conjugate compete for the anti-AGRIN antibody binding site. Since the number of sites is limited, as more sites are occupied by AGRIN from the sample, fewer sites are left to bind AGRIN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AGRIN concentration in each sample is interpolated from this standard curve.

NCBI Summary for Agrin
This gene encodes one of several proteins that are critical in the development of the neuromuscular junction (NMJ), as identified in mouse knock-out studies. The encoded protein contains several laminin G, Kazal type serine protease inhibitor, and epidermal growth factor domains. Additional post-translational modifications occur to add glycosaminoglycans and disulfide bonds. In one family with congenital myasthenic syndrome affecting limb-girdle muscles, a mutation in this gene was found. [provided by RefSeq, Aug 2011]
UniProt Comments for Agrin
AGRN: Plays a central role in the formation and the maintenance of the neuromuscular junction (NMJ), the synapse between motor neuron and skeletal muscle. Ligand of the MUSK signaling complex that directly binds LRP4 in this complex and induces the phosphorylation of MUSK, the kinase of the complex. The activation of MUSK in myotubes induces the formation of NMJ by regulating different processes including the transcription of specific genes and the clustering of AChR in the postsynaptic membrane. Defects in AGRN are a cause of myasthenia, limb-girdle, familial (LGM). A congenital myasthenic syndrome characterized by a typical 'limb girdle' pattern of muscle weakness with small, simplified neuromuscular junctions but normal acetylcholine receptor and acetylcholinesterase function.
