The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing AKR1C3. The ELISA analytical biochemical technique of the MBS7220102 kit is based on AKR1C3 antibody-AKR1C3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect AKR1C3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, AKR1C3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: AKR1C3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AKR1C3 antibody and an AKR1C3-HRP conjugate. The assay sample and buffer are incubated together with AKR1C3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AKR1C3 concentration since AKR1C3 from samples and AKR1C3-HRP conjugate compete for the anti-AKR1C3 antibody binding site. Since the number of sites is limited, as more sites are occupied by AKR1C3 from the sample, fewer sites are left to bind AKR1C3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AKR1C3 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the aldo/keto reductase superfamily, which consists of more than 40 known enzymes and proteins. These enzymes catalyze the conversion of aldehydes and ketones to their corresponding alcohols by utilizing NADH and/or NADPH as cofactors. The enzymes display overlapping but distinct substrate specificity. This enzyme catalyzes the reduction of prostaglandin (PG) D2, PGH2 and phenanthrenequinone (PQ), and the oxidation of 9alpha,11beta-PGF2 to PGD2. It may play an important role in the pathogenesis of allergic diseases such as asthma, and may also have a role in controlling cell growth and/or differentiation. This gene shares high sequence identity with three other gene members and is clustered with those three genes at chromosome 10p15-p14.
