The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing AMACR. The ELISA analytical biochemical technique of the MBS901785 kit is based on AMACR antibody-AMACR antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect AMACR antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, AMACR. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for AMACR has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any AMACR present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for AMACR is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AMACR bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a racemase. The encoded enzyme interconverts pristanoyl-CoA and C27-bile acylCoAs between their (R)- and (S)-stereoisomers. The conversion to the (S)-stereoisomers is necessary for degradation of these substrates by peroxisomal beta-oxidation. Encoded proteins from this locus localize to both mitochondria and peroxisomes. Mutations in this gene may be associated with adult-onset sensorimotor neuropathy, pigmentary retinopathy, and adrenomyeloneuropathy due to defects in bile acid synthesis. Alternatively spliced transcript variants have been described. Read-through transcription also exists between this gene and the upstream neighboring C1QTNF3 (C1q and tumor necrosis factor related protein 3) gene. [provided by RefSeq, Mar 2011]
