The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ANP. The ELISA analytical biochemical technique of the MBS723537 kit is based on ANP antibody-ANP antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ANP antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ANP. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Intended Uses: This ANP ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ANP. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: ANP ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ANP antibody and an ANP-HRP conjugate. The assay sample and buffer are incubated together with ANP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ANP concentration since ANP from samples and ANP-HRP conjugate compete for the anti-ANP antibody binding site. Since the number of sites is limited, as more sites are occupied by ANP from the sample, fewer sites are left to bind ANP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ANP concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene belongs to the natriuretic peptide family. Natriuretic peptides are implicated in the control of extracellular fluid volume and electrolyte homeostasis. This protein is synthesized as a large precursor (containing a signal peptide), which is processed to release a peptide from the N-terminus with similarity to vasoactive peptide, cardiodilatin, and another peptide from the C-terminus with natriuretic-diuretic activity. Mutations in this gene have been associated with atrial fibrillation familial type 6. [provided by RefSeq, Sep 2009]
