MBS705428 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the apelin 36 (AP36) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing AP36. The ELISA analytical biochemical technique of the MBS705428 kit is based on AP36 antibody-AP36 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect AP36 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, AP36. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Introduction: The apelin gene encodes a preproprotein that is processed to generate a variety of bioactive peptides, including those having 36, 17, or 13 amino acids (apelin-36, apelin-17, and apelin-13, respectively). The apelin proteins are the endogenous ligands of the G protein-coupled receptor, APJ. Apelin-36 is the full-length mature peptide produced from the translated 77 amino acid prepropeptide. The apelins act primarily in the peripheral and central nervous system, playing important roles in regulating cardiovascular function, fluid homeostasis, hypertension, and insulin sensitivity. Apelin-36 is a less potent agonist of APJ than either apelin-17 or apelin-13 (EC50 = 20, 2.5, and 0.37 nM, respectively). Apelin-36 potently inhibits HIV-1 entry into cells expressing APJ and CD4, limiting HIV infection.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to AP36. Standards or samples are then added to the appropriate microtiter plate wells with a Horseradish Peroxidase (HRP)-conjugated antibody preparation specific for AP36 and incubated. Then substrate solution A and B are added to each well. Only those wells that contain AP36, HRP-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of AP36 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
