The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing APOB. The ELISA analytical biochemical technique of the MBS701268 kit is based on APOB antibody-APOB antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect APOB antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, APOB. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to APO-B100. Standards or samples are then added to the appropriate microtiter plate wells with a biotin conjugated polyclonal antibody preparation specific for APO-B100 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain APO-B100, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of APO-B100 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene product is the main apolipoprotein of chylomicrons and low density lipoproteins. It occurs in plasma as two main isoforms, apoB-48 and apoB-100: the former is synthesized exclusively in the gut and the latter in the liver. The intestinal and the hepatic forms of apoB are encoded by a single gene from a single, very long mRNA. The two isoforms share a common N-terminal sequence. The shorter apoB-48 protein is produced after RNA editing of the apoB-100 transcript at residue 2180 (CAA->UAA), resulting in the creation of a stop codon, and early translation termination. Mutations in this gene or its regulatory region cause hypobetalipoproteinemia, normotriglyceridemic hypobetalipoproteinemia, and hypercholesterolemia due to ligand-defective apoB, diseases affecting plasma cholesterol and apoB levels.
