The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing Bax. The ELISA analytical biochemical technique of the MBS2023569 kit is based on Bax antibody-Bax antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Bax antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, Bax. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Bcl2 Associated X Protein (Bax). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Bcl2 Associated X Protein (Bax). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Bcl2 Associated X Protein (Bax), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Bcl2 Associated X Protein (Bax) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene belongs to the BCL2 protein family. BCL2 family members form hetero- or homodimers and act as anti- or pro-apoptotic regulators that are involved in a wide variety of cellular activities. This protein forms a heterodimer with BCL2, and functions as an apoptotic activator. This protein is reported to interact with, and increase the opening of, the mitochondrial voltage-dependent anion channel (VDAC), which leads to the loss in membrane potential and the release of cytochrome c. The expression of this gene is regulated by the tumor suppressor P53 and has been shown to be involved in P53-mediated apoptosis.
