The ELISA analytical biochemical technique of the MBS7207186 kit is based on BOLA3 antibody-BOLA3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect BOLA3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, BOLA3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the assay: BOLA3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-BOLA3 antibody and an BOLA3-HRP conjugate. The assay sample and buffer are incubated together with BOLA3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BOLA3 concentration since BOLA3 from samples and BOLA3-HRP conjugate compete for the anti-BOLA3 antibody binding site. Since the number of sites is limited, as more sites are occupied by BOLA3 from the sample, fewer sites are left to bind BOLA3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BOLA3 concentration in each sample is interpolated from this standard curve.

This gene encodes a protein that plays an essential role in the production of iron-sulfur (Fe-S) clusters for the normal maturation of lipoate-containing 2-oxoacid dehydrogenases, and for the assembly of the mitochondrial respiratory chain complexes. Mutation in this gene has been associated with multiple mitochondrial dysfunctions syndrome-2. Two alternatively spliced transcript variants encoding different isoforms with distinct subcellular localization have been reported for this gene (PMID:21944046). [provided by RefSeq, Dec 2011]
