MBS916031 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the biotinidase (BTD) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing BTD. The ELISA analytical biochemical technique of the MBS916031 kit is based on BTD antibody-BTD antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect BTD antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, BTD. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to BTD. Standards or samples are added to the appropriate microtiter plate wells with Horseradish Peroxidase (HRP) conjugated BTD. The competitive inhibition reaction is launched between with HRP-conjugated BTD and BTD in samples. A substrate solution is added to the wells and the color develops in opposite to the amount of BTD in the sample. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene functions to recycle protein-bound biotin by cleaving biocytin (biotin-epsilon-lysine), a normal product of carboxylase degradation, resulting in regeneration of free biotin. The encoded protein has also been shown to have biotinyl transferase activity. Mutations in this gene are associated with biotinidase deficiency. Multiple transcript variants encoding different isoforms have been described. [provided by RefSeq, Aug 2013]

Defects in BTD are the cause of biotinidase deficiency (BTD deficiency); also called late-onset multiple carboxylase deficiency. BTD deficiency is a juvenile form of multiple carboxylase deficiency, an autosomal recessive disorder of biotin metabolism, characterized by ketoacidosis, hyperammonemia, excretion of abnormal organic acid metabolites, and dermatitis. BTD deficiency is characterized by seizures, hypotonia, skin rash, alopecia, ataxia, hearing loss, and optic atrophy.
