MBS939954 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the complement component 9 (C9) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing C9. The ELISA analytical biochemical technique of the MBS939954 kit is based on C9 antibody-C9 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect C9 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, C9. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for C9 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any C9 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for C9 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of C9 bound in the initial step. The color development is stopped and the intensity of the color is measured.

UniProt Comments for C9
C9: Constituent of the membrane attack complex (MAC) that plays a key role in the innate and adaptive immune response by forming pores in the plasma membrane of target cells. C9 is the pore-forming subunit of the MAC. Defects in C9 are a cause of complement component 9 deficiency (C9D). A rare defect of the complement classical pathway associated with susceptibility to severe recurrent infections, predominantly by Neisseria gonorrhoeae or Neisseria meningitidis. Belongs to the complement C6/C7/C8/C9 family.

Protein type: Extracellular matrix; Membrane protein, multi-pass; Membrane protein, integral; Apoptosis

Cellular Component: membrane attack complex; membrane; cytoplasm; integral to membrane; extracellular region; plasma membrane

Biological Process: immune system process; cytolysis; complement activation, alternative pathway; innate immune response; immune response; blood coagulation; complement activation, classical pathway
