Calcineurin plays an important role in the T-cell receptor-mediated signal transduction pathway. The protein encoded by this gene binds specifically to the activated form of calcineurin and inhibits calcineurin-mediated signal transduction. The encoded protein is found in the nucleus and contains a leucine zipper domain as well as several PEST motifs, sequences which confer targeted degradation to those proteins which contain them. Alternative splicing results in multiple transcript variants encoding two different isoforms. [provided by RefSeq, Jan 2011]

CABIN1: an inhibitor of the calcium-dependent serine/threonine phosphatase, calcineurin. Inhibits calcineurin signaling in T cells, regulating T cell receptor (TCR) signaling, transcription, and apoptosis. Inhibition of activated calcineurin is dependent on both PKC and calcium signals. Represses myocyte enhancer factor 2 (MEF2)-mediated transcription by recruiting chromatin remodeling enzymes, and also negatively regulates the activity of the tumor suppressor p53. Is degraded in response to genotoxic stress, releasing its inhibition of p53, allowing p53 to elicit cellular stress responses. Involved in regulation of chromatin structure as part of the highly conserved HIRA/UBN1/CABIN1/ASF1A (HUCA) histone chaperone complex.
