The ELISA analytical biochemical technique of the MBS7229003 kit is based on CARD10 antibody-CARD10 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CARD10 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CARD10. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Intended Uses: This CARD10 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CARD10. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: CARD10 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CARD10 antibody and an CARD10-HRP conjugate. The assay sample and buffer are incubated together with CARD10-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CARD10 concentration since CARD10 from samples and CARD10-HRP conjugate compete for the anti-CARD10 antibody binding site. Since the number of sites is limited, as more sites are occupied by CARD10 from the sample, fewer sites are left to bind CARD10-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CARD10 concentration in each sample is interpolated from this standard curve.

The caspase recruitment domain (CARD) is a protein module that consists of 6 or 7 antiparallel alpha helices. It participates in apoptosis signaling through highly specific protein-protein homophilic interactions. Like several other CARD proteins, CARD10 belongs to the membrane-associated guanylate kinase (MAGUK) family and activates NF-kappa-B (NFKB; see MIM 164011) through BCL10 (MIM 603517) (Wang et al., 2001 [PubMed 11259443]).[supplied by OMIM, Mar 2008]

CARD proteins function as scaffolds for the assembly of multiprotein complexes at specialized regions of the plasma membrane. The CARD domain of CARD14 associates specifically with the CARD domains of CARD10 and BCL10, a signaling protein that activates NF-kB through the IkappaB kinase complex in response to upstream stimuli. When expressed in cells, CARD14 activates NF-kappaB and induces the phosphorylation of BCL10. A subgroup of primary lymphomas of the central nervous system have higher levels of CARD14 expression.
