MBS288535 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Caspase-6 (Casp6) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing Casp6. The ELISA analytical biochemical technique of the MBS288535 kit is based on Casp6 antibody-Casp6 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Casp6 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, Casp6. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to CASP-6. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for CASP-6 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain CASP-6, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of CASP-6 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Summary for Casp6
member of the cysteine-aspartic acid protease (caspase) family that plays a terminal role in apoptosis [RGD, Feb 2006]
UniProt Comments for Casp6
CASP6: Involved in the activation cascade of caspases responsible for apoptosis execution. Cleaves poly(ADP-ribose) polymerase in vitro, as well as lamins. Overexpression promotes programmed cell death. Heterotetramer that consists of two anti-parallel arranged heterodimers, each one formed by a 18 kDa (p18) and a 11 kDa (p11) subunit. Interacts with BIRC6/bruce. Activation is suppressed by phosphorylation at Ser-257. Belongs to the peptidase C14A family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: EC 3.4.22.59; Protease; Apoptosis

Cellular Component: cytoplasm; cytosol; nucleoplasm; nucleus

Molecular Function: cysteine-type endopeptidase activity; endopeptidase activity; identical protein binding

Biological Process: acute inflammatory response to non-antigenic stimulus; apoptosis; epithelial cell differentiation; lens development in camera-type eye; positive regulation of apoptosis; positive regulation of neuron apoptosis; positive regulation of retinal cell programmed cell death; proteolysis; response to cytokine stimulus; response to estradiol stimulus; response to gamma radiation; response to glucose stimulus; response to heat; response to hydrogen peroxide; response to iron ion; response to metal ion; response to organic nitrogen; response to organic substance; response to testosterone stimulus
