The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CDH10. The ELISA analytical biochemical technique of the MBS7201919 kit is based on CDH10 antibody-CDH10 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CDH10 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CDH10. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: CDH10 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CDH10 antibody and an CDH10-HRP conjugate. The assay sample and buffer are incubated together with CDH10-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CDH10 concentration since CDH10 from samples and CDH10-HRP conjugate compete for the anti-CDH10 antibody binding site. Since the number of sites is limited, as more sites are occupied by CDH10 from the sample, fewer sites are left to bind CDH10-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CDH10 concentration in each sample is interpolated from this standard curve.

This gene encodes a type II classical cadherin from the cadherin superfamily, integral membrane proteins that mediate calcium-dependent cell-cell adhesion. Mature cadherin proteins are composed of a large N-terminal extracellular domain, a single membrane-spanning domain, and a small, highly conserved C-terminal cytoplasmic domain. The extracellular domain consists of 5 subdomains, each containing a cadherin motif, and appears to determine the specificity of the protein's homophilic cell adhesion activity. Type II (atypical) cadherins are defined based on their lack of a HAV cell adhesion recognition sequence specific to type I cadherins. This particular cadherin is predominantly expressed in brain and is putatively involved in synaptic adhesions, axon outgrowth and guidance. [provided by RefSeq, Jan 2011]
