MBS724080 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Choline acetyltransferase (ChAT) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ChAT. The ELISA analytical biochemical technique of the MBS724080 kit is based on ChAT antibody-ChAT antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ChAT antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ChAT. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This ChAT ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse ChAT.

Principle of the Assay: ChAT ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ChAT antibody and an ChAT-HRP conjugate. The assay sample and buffer are incubated together with ChAT-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ChAT concentration since ChAT from samples and ChAT-HRP conjugate compete for the anti-ChAT antibody binding site. Since the number of sites is limited, as more sites are occupied by ChAT from the sample, fewer sites are left to bind ChAT-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ChAT concentration in each sample is interpolated from this standard curve.

This gene encodes an enzyme which catalyzes the biosynthesis of the neurotransmitter acetylcholine. This gene product is a characteristic feature of cholinergic neurons, and changes in these neurons may explain some of the symptoms of Alzheimer's disease. Polymorphisms in this gene have been associated with Alzheimer's disease and mild cognitive impairment. Mutations in this gene are associated with congenital myasthenic syndrome associated with episodic apnea. Multiple transcript variants encoding different isoforms have been found for this gene, and some of these variants have been shown to encode more than one isoform. [provided by RefSeq, May 2010]
