Species Reactivity
Specificity
This assay has high sensitivity and excellent specificity for detection of CLDN-1. No significant cross-reactivity or interference between CLDN-1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CLDN-1 and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate

Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of CLDN1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS724224 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Claudin 1 (CLDN1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CLDN1. The ELISA analytical biochemical technique of the MBS724224 kit is based on CLDN1 antibody-CLDN1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CLDN1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CLDN1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This CLDN-1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse CLDN-1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay||CLDN-1 ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for CLDN-1. Standards or samples are then added to the microtiter plate wells and CLDN-1 if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of CLDN-1 present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for CLDN-1 are added to each well to "sandwich" the CLDN-1 immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain CLDN-1 and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLDN-1 concentration in each sample is interpolated from this standard curve.

NCBI Summary for CLDN1
Tight junctions represent one mode of cell-to-cell adhesion in epithelial or endothelial cell sheets, forming continuous seals around cells and serving as a physical barrier to prevent solutes and water from passing freely through the paracellular space. These junctions are comprised of sets of continuous networking strands in the outwardly facing cytoplasmic leaflet, with complementary grooves in the inwardly facing extracytoplasmic leaflet. The protein encoded by this gene, a member of the claudin family, is an integral membrane protein and a component of tight junction strands. Loss of function mutations result in neonatal ichthyosis-sclerosing cholangitis syndrome. [provided by RefSeq, Jul 2008]
UniProt Comments for CLDN1
Claudin-1: Plays a major role in tight junction-specific obliteration of the intercellular space, through calcium- independent cell-adhesion activity. Acts as a co- receptor for HCV entry into hepatic cells. Can form homo- and heteropolymers with other CLDN. Homopolymers interact with CLDN3, but not CLDN2, homopolymers. Directly interacts with TJP1/ZO-1, TJP2/ZO-2 and TJP3/ZO-3. Interacts with MPDZ and INADL. May interact with HCV E1 and E2 proteins. Strongly expressed in liver and kidney. Expressed in heart, brain, spleen, lung and testis. Belongs to the claudin family.
