Intended Uses: This CSHL1ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse CSHL1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: CSHL1ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CSHL1antibody and an CSHL1-HRP conjugate. The assay sample and buffer are incubated together with CSHL1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CSHL1concentration since CSHL1from samples and CSHL1-HRP conjugate compete for the anti-CSHL1antibody binding site. Since the number of sites is limited, as more sites are occupied by CSHL1from the sample, fewer sites are left to bind CSHL1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CSHL1concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the somatotropin/prolactin family of hormones which play an important role in growth control. The gene, along with four other related genes, is located at the growth hormone locus on chromosome 17 where they are interspersed in the same transcriptional orientation; an arrangement which is thought to have evolved by a series of gene duplications. Although the five genes share a remarkably high degree of sequence identity, they are expressed selectively in different tissues. This particular family member is expressed in placental villi, although it was originally thought to be a pseudogene. In fact, alternative splicing suggests that the majority of the transcripts would be unable to express a secreted protein. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Jul 2008]
