MBS176488 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the CXCL13/BLC, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CXCL13. The ELISA analytical biochemical technique of the MBS176488 kit is based on CXCL13 antibody-CXCL13 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CXCL13 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CXCL13. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: human CXCL13 ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for CXCL13 has been precoated onto 96-well plates. Standards (E.coli,V23-R94) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CXCL13 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human CXCL13 amount of sample captured in plate.
Background: C-X-C motif chemokine 13 (CXCL13), also known as B lymphocyte chemoattractant (BLC), is a protein that in humans is encoded by theCXCL13 gene. CXCL13 is a small cytokine belonging to the CXC chemokine family, and it was located on sequence maps to 4q21, near the genes encoding most CXC chemokines. In T-lymphocytes, CXCL13 expression is thought to reflect a germinal center origin of the T-cell. Hence, expression of CXCL13 in T-cell lymphomas, such as Angioimmunoblastic T-cell Lymphoma, is thought to reflect a germinal center origin of the neoplastic T-cells

NCBI Summary for CXCL13
B lymphocyte chemoattractant, independently cloned and named Angie, is an antimicrobial peptide and CXC chemokine strongly expressed in the follicles of the spleen, lymph nodes, and Peyer's patches. It preferentially promotes the migration of B lymphocytes (compared to T cells and macrophages), apparently by stimulating calcium influx into, and chemotaxis of, cells expressing Burkitt's lymphoma receptor 1 (BLR-1). It may therefore function in the homing of B lymphocytes to follicles. [provided by RefSeq, Oct 2014]
UniProt Comments for CXCL13
CXCL13: Chemotactic for B-lymphocytes but not for T-lymphocytes, monocytes and neutrophils. Does not induce calcium release in B- lymphocytes. Binds to BLR1/CXCR5. Belongs to the intercrine alpha (chemokine CxC) family.
