This assay has high sensitivity and excellent specificity for detection of CXCL-17. No significant cross-reactivity or interference between CXCL-17 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CXCL-17 and all the analogues, therefore, cross reaction may still exist in some cases.

The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CXCL17. The ELISA analytical biochemical technique of the MBS7239012 kit is based on CXCL17 antibody-CXCL17 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CXCL17 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CXCL17. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This CXCL-17 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CXCL-17. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: CXCL-17 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CXCL-17 antibody and an CXCL-17-HRP conjugate. The assay sample and buffer are incubated together with CXCL-17-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CXCL-17 concentration since CXCL-17 from samples and CXCL-17-HRP conjugate compete for the anti-CXCL-17 antibody binding site. Since the number of sites is limited, as more sites are occupied by CXCL-17 from the sample, fewer sites are left to bind CXCL-17-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CXCL-17 concentration in each sample is interpolated from this standard curve.

This gene is located in a cluster of similar genes in the same region of chromosome 17. These genes encode CC cytokines, which are secreted proteins characterized by two adjacent cysteines. The product of this gene is chemotactic for T cells and monocytes, and acts through C-C chemokine receptor type 1 (CCR1). The proprotein is further processed into numerous smaller functional peptides. Naturally-occurring readthrough transcripts occur from this gene into the downstream gene, CCL14 (chemokine (C-C motif) ligand 14). [provided by RefSeq, Jan 2013]
