The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest human 17-alpha-OH concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.

The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CYP17A1. The ELISA analytical biochemical technique of the MBS702060 kit is based on CYP17A1 antibody-CYP17A1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CYP17A1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CYP17A1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with goat-anti-rabbit antibody. Standards or samples are added to the appropriate microtiter plate wells with an antibody specific for 17-alpha-OH and Horseradish Peroxidase (HRP) conjugated 17-alpha-OH. The competitive inhibition reaction is launched between with HRP labeled 17-alpha-OH and unlabeled 17-alpha-OH with the antibody. A substrate solution is added to the wells and the color develops in opposite to the amount of 17-alpha-OH in the sample. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum. It has both 17alpha-hydroxylase and 17,20-lyase activities and is a key enzyme in the steroidogenic pathway that produces progestins, mineralocorticoids, glucocorticoids, androgens, and estrogens. Mutations in this gene are associated with isolated steroid-17 alpha-hydroxylase deficiency, 17-alpha-hydroxylase/17,20-lyase deficiency, pseudohermaphroditism, and adrenal hyperplasia.
