The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing DDX23. The ELISA analytical biochemical technique of the MBS281839 kit is based on DDX23 antibody-DDX23 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect DDX23 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, DDX23. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate DDX23 in samples. An antibody specific for DDX23 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDDX23 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DDX23 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DDX23 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the DEAD box protein family. DEAD box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD), are putative RNA helicases. They are implicated in a number of cellular processes involving alteration of RNA secondary structure, such as translation initiation, nuclear and mitochondrial splicing, and ribosome and spliceosome assembly. Based on their distribution patterns, some members of this family are believed to be involved in embryogenesis, spermatogenesis, and cellular growth and division. The protein encoded by this gene is a component of the U5 snRNP complex; it may facilitate conformational changes in the spliceosome during nuclear pre-mRNA splicing. An alternatively spliced transcript variant has been found for this gene, but its biological validity has not been determined. [provided by RefSeq, Jul 2008]
