Species Reactivity
Specificity
This assay has high sensitivity and excellent specificity for detection of bovine DPP4. No significant cross-reactivity or interference between bovine DPP4 and analogues was observed.
Samples
Serum, plasma, tissue homogenates
Assay Type
Competitive
Detection Range
0.312 ng/ml -20 ng/ml.
Sensitivity
The minimum detectable dose of bovine DPP4 is typically less than 0.156 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest bovine DPP4 concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision
Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision
Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
Preparation and Storage
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
ISO Certification
Manufactured in an ISO 9001:2008 Certified Laboratory.
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of DPP4 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS932554 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the dipeptidyl-peptidase 4 (DPP4) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing DPP4. The ELISA analytical biochemical technique of the MBS932554 kit is based on DPP4 antibody-DPP4 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect DPP4 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, DPP4. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with DPP4. Standards or samples are added to the appropriate microtiter plate wells with Horseradish Peroxidase (HRP) conjugated antibody preparation specific for DPP4. The competitive inhibition reaction is launched between with pre-coated DPP4 and DPP4 in samples. A substrate solution is added to the wells and the color develops in opposite to the amount of DPP4 in the sample. The color development is stopped and the intensity of the color is measured.

Function: Cell surface glycoprotein receptor involved in the costimulatory signal essential for T-cell receptor (TCR)-mediated T-cell activation. Acts as a positive regulator of T-cell coactivation, by binding at least ADA, CAV1, IGF2R, and PTPRC. Its binding to CAV1 and CARD11 induces T-cell proliferation and NF-kappa-B activation in a T-cell receptor/CD3-dependent manner. Its interaction with ADA also regulates lymphocyte-epithelial cell adhesion. In association with FAP is involved in the pericellular proteolysis of the extracellular matrix (ECM), the migration and invasion of endothelial cells into the ECM. May be involved in the promotion of lymphatic endothelial cells adhesion, migration and tube formation. When overexpressed, enhanced cell proliferation, a process inhibited by GPC3. Acts also as a serine exopeptidase with a dipeptidyl peptidase activity that regulates various physiological processes by cleaving peptides in the circulation, including many chemokines, mitogenic growth factors, neuropeptides and peptide hormones. Removes N-terminal dipeptides sequentially from polypeptides having unsubstituted N-termini provided that the penultimate residue is proline

Catalytic activity: Release of an N-terminal dipeptide, Xaa-Yaa-|-Zaa-, from a polypeptide, preferentially when Yaa is Pro, provided Zaa is neither Pro nor hydroxyproline.

Enzyme regulation: Inhibited by GPC3 and diprotin A
