MBS2503584 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the EPCAM, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing EPCAM. The ELISA analytical biochemical technique of the MBS2503584 kit is based on EPCAM antibody-EPCAM antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect EPCAM antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, EPCAM. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Mouse EPCAM concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to EPCAM. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for EPCAM and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain EPCAM, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of EPCAM. You can calculate the concentration of EPCAM in the samples by comparing the OD of the samples to the standard curve.

NCBI Summary for EPCAM
This gene encodes a carcinoma-associated antigen and is a member of a family that includes at least two type I membrane proteins. This antigen is expressed on most normal epithelial cells and gastrointestinal carcinomas and functions as a homotypic calcium-independent cell adhesion molecule. The antigen is being used as a target for immunotherapy treatment of human carcinomas. Mutations in this gene result in congenital tufting enteropathy. [provided by RefSeq, Dec 2008]
UniProt Comments for EPCAM
EPCAM: a single-pass type I membrane protein of the EPCAM family. May act as a physical homophilic interaction molecule between intestinal epithelial cells (IECs) and intraepithelial lymphocytes (IELs) at the mucosal epithelium for providing immunological barrier as a first line of defense against mucosal infection. Expressed in almost all epithelial cell membranes but not on mesodermal or neural cell membranes. Found on the surface of adenocarcinomas including pancreatic carcinoma. May be a marker of cancer stem cells.

Protein type: Motility/polarity/chemotaxis; Membrane protein, integral

Chromosomal Location of Human Ortholog: 2p21

Cellular Component: tight junction; cell surface; basolateral plasma membrane; apical plasma membrane; integral to membrane; plasma membrane; lateral plasma membrane

Molecular Function: protein binding; protein complex binding

Biological Process: ureteric bud development; positive regulation of cell proliferation; positive regulation of transcription from RNA polymerase II promoter; stem cell differentiation

Disease: Diarrhea 5, With Tufting Enteropathy, Congenital; Colorectal Cancer, Hereditary Nonpolyposis, Type 8
