Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of EPX elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS2024555 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Eosinophil Peroxidase (EPX) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing EPX. The ELISA analytical biochemical technique of the MBS2024555 kit is based on EPX antibody-EPX antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect EPX antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, EPX. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

EPX elisa kit
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Eosinophil Peroxidase (EPX). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Eosinophil Peroxidase (EPX). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Eosinophil Peroxidase (EPX), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Eosinophil Peroxidase (EPX) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Summary for EPX
This gene is a member of the peroxidase gene family and is expressed in eosinophils. The encoded precursor protein is processed into covalently attached heavy and light chains to form the mature enzyme, which functions as an oxidant. The enzyme is released at sites of parasitic infection or allergen stimulation to mediate lysis of protozoa or parasitic worms. The gene is found in a cluster of three peroxidase genes at chromosome 17q23. Mutations in this gene result in eosinophil peroxidase deficiency. [provided by RefSeq, Sep 2009]
UniProt Comments for EPX
EPX: Mediates tyrosine nitration of secondary granule proteins in mature resting eosinophils. Shows significant inhibitory activity towards Mycobacterium tuberculosis H37Rv by inducing bacterial fragmentation and lysis. Defects in EPX are the cause of eosinophil peroxidase deficiency (EPD). EPD is an autosomal recessive defect where anomalous eosinophils are characterized by nuclear hypersegmentation, hypogranulation, and negative peroxidase and phospholipid staining. Belongs to the peroxidase family. XPO subfamily.

Protein type: Oxidoreductase; EC 1.11.1.7

Chromosomal Location of Human Ortholog: 17q23.1

Molecular Function: peroxidase activity; metal ion binding; heme binding

Biological Process: positive regulation of interleukin-4 production; negative regulation of interleukin-10 production; hydrogen peroxide catabolic process; negative regulation of interleukin-5 production; response to oxidative stress; defense response to nematode

Disease: Eosinophil Peroxidase Deficiency
