The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ERBB4. The ELISA analytical biochemical technique of the MBS915273 kit is based on ERBB4 antibody-ERBB4 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ERBB4 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ERBB4. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ERBB4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ERBB4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ERBB4 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ERBB4 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is a member of the Tyr protein kinase family and the epidermal growth factor receptor subfamily. It encodes a single-pass type I membrane protein with multiple cysteine rich domains, a transmembrane domain, a tyrosine kinase domain, a phosphotidylinositol-3 kinase binding site and a PDZ domain binding motif. The protein binds to and is activated by neuregulins and other factors and induces a variety of cellular responses including mitogenesis and differentiation. Multiple proteolytic events allow for the release of a cytoplasmic fragment and an extracellular fragment. Mutations in this gene have been associated with cancer. Alternatively spliced variants which encode different protein isoforms have been described; however, not all variants have been fully characterized. [provided by RefSeq, Jul 2008]
