The ELISA analytical biochemical technique of the MBS701478 kit is based on ERCC1 antibody-ERCC1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ERCC1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ERCC1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ERCC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ERCC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ERCC1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ERCC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The product of this gene functions in the nucleotide excision repair pathway, and is required for the repair of DNA lesions such as those induced by UV light or formed by electrophilic compounds including cisplatin. The encoded protein forms a heterodimer with the XPF endonuclease (also known as ERCC4), and the heterodimeric endonuclease catalyzes the 5' incision in the process of excising the DNA lesion. The heterodimeric endonuclease is also involved in recombinational DNA repair and in the repair of inter-strand crosslinks. Mutations in this gene result in cerebrooculofacioskeletal syndrome, and polymorphisms that alter expression of this gene may play a role in carcinogenesis. Multiple transcript variants encoding different isoforms have been found for this gene.
