The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing FGF9. The ELISA analytical biochemical technique of the MBS176006 kit is based on FGF9 antibody-FGF9 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect FGF9 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, FGF9. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: human FGF9 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for FGF9 has been precoated onto 96-well plates. Standards(sf21,M1-S208) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for FGF9 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human FGF9 amount of sample captured in plate.
Background: Fibroblast growth factor-9 (FGF-9) is a steroid-regulated mitogen and survival factor for nerve and mesenchymal cells.1 The human FGF-9 cDNA cloned by using oligonucleotide probes encodes a polypeptide consisting of 208amino acids. Sequence similarity to other members of the FGF family has been estimated to be around 30%.2FGF-9 is an autocrine estromedin endometrial stromal growth factor that plays roles in cyclic proliferation of uterine endometrial stroma.3 FGF9 is produced and secreted by the prostatic stromal cells. It is a potent mitogen for bothprostatic epithelial and stromal cells in culture. FGF9 is an abundant secreted growth factor that can act as both a paracrine mitogen for epithelial cells and an autocrine mitogen for stromal cells. Overexpression of this paracrine and autocrine growth factor may play an important role in the epithelial and stromal proliferation in benign prostatic hyperplasia.4 As a result of glycosylation, the molecular mass is 25-27KDa.

The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. This protein was isolated as a secreted factor that exhibits a growth-stimulating effect on cultured glial cells. In nervous system, this protein is produced mainly by neurons and may be important for glial cell development. Expression of the mouse homolog of this gene was found to be dependent on Sonic hedgehog (Shh) signaling. Mice lacking the homolog gene displayed a male-to-female sex reversal phenotype, which suggested a role in testicular embryogenesis. [provided by RefSeq, Jul 2008]
