The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing FTH1. The ELISA analytical biochemical technique of the MBS931412 kit is based on FTH1 antibody-FTH1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect FTH1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, FTH1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for FTH1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FTH1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FTH1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FTH1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Ferritin-H: Stores iron in a soluble, non-toxic, readily available form. Important for iron homeostasis. Has ferroxidase activity. Iron is taken up in the ferrous form and deposited as ferric hydroxides after oxidation. Also plays a role in delivery of iron to cells. Mediates iron uptake in capsule cells of the developing kidney. Oligomer of 24 subunits. There are two types of subunits: L (light) chain and H (heavy) chain. The major chain can be light or heavy, depending on the species and tissue type. The functional molecule forms a roughly spherical shell with a diameter of 12 nm and contains a central cavity into which the insoluble mineral iron core is deposited. Belongs to the ferritin family.
