The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing gamma-Secretase. The ELISA analytical biochemical technique of the MBS2516301 kit is based on gamma-Secretase antibody-gamma-Secretase antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect gamma-Secretase antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, gamma-Secretase. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This ELISA kit can be applied to the in vitro quantitative determination of Rat ?-Secretase concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been precoated with an antibody specific to Rat ?-Secretase . Standards or samples are added to appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibodies specific for Rat ?-Secretase and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well successively and incubated. After incubation, free components are washed away. Then the Substrate Reagent is added to each well, only those wells that contain Rat ?-Secretase , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction will be terminated by adding Stop Solution and appears yellow in color. The optical density (OD) can be measured with spectrophotometry at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Rat ?-Secretase . The concentration of Rat ?-Secretase in samples can be calculated by comparing the OD of the samples with the standard curve.

Presenilins, which are components of the gamma-secretase protein complex, are required for intramembranous processing of some type I transmembrane proteins, such as the Notch proteins and the beta-amyloid precursor protein. Signaling by Notch receptors mediates a wide range of developmental cell fates. Processing of the beta-amyloid precursor protein generates neurotoxic amyloid beta peptides, the major component of senile plaques associated with Alzheimer's disease. This gene encodes a protein that is required for Notch pathway signaling, and for the activity and accumulation of gamma-secretase. Mutations resulting in haploinsufficiency for this gene cause familial acne inversa-2 (ACNINV2). Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2013]
