The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing HIST1H3A. The ELISA analytical biochemical technique of the MBS937951 kit is based on HIST1H3A antibody-HIST1H3A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect HIST1H3A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, HIST1H3A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for HIST1H3A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HIST1H3A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HIST1H3A is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HIST1H3A bound in the initial step. The color development is stopped and the intensity of the color is measured.

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. This structure consists of approximately 146 bp of DNA wrapped around a nucleosome, an octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a member of the histone H3 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6p22-p21.3. [provided by RefSeq, Jul 2008]
