MBS267094 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the leukocyte antigen G (HLA-G) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing HLA-G. The ELISA analytical biochemical technique of the MBS267094 kit is based on HLA-G antibody-HLA-G antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect HLA-G antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, HLA-G. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%). Human leukocyte antigen G exists as a cell membrane localized and soluble human form. As a clinical hum immunol biomarker for placental development, research interest in expression and localization is an active area. Aiding these studies is an HLAG ELISA kit suitable for analysis of cells in culture, tissue homogenates and other samples requiring a broad detection range, including soluble HLA G. These HLAG ELISA kits are designed as double-sandwich format, unlike competitor products category members when ELISA kit select compare assay products is chosen. Adding to the antigen HLA kit 1 value are included kit 1 plate with HLA antibody 5a6g7, anti mouse IgG1 biotin secondary antibodies, standard protein product and either Alexa 174 fluor or AMP detection as described in plate datasheet. To purchase HLA antigen ELISA kit 1, to detect soluble human leukocyte antigen G, in the United States or other Republic countries and Islands, see MyBiosource.com

Principle of the Assay: This experiment use double-sandwich elisa technique and the ELISA Kit provided is typical. The pre-coated antibody is human HLA-G monoclonal antibody and the detecting antibody is polyclonal antibody with biotin labeled. Samples and biotin labeling antibody are added into ELISA plate wells and washed out with PBS or TBS. Then Avidin-peroxidase conjugates are added to ELISA wells in order; Use TMB substrate for coloring after reactant thoroughly washed out by PBS or TBS. TMB turns into blue in peroxidase catalytic and finally turns into yellow under the action of acid. The color depth and the testing factors in samples are positively correlated.

NCBI Summary for HLA-G
HLA-A belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45 kDa and its gene contains 8 exons. Exon 1 encodes the leader peptide, exons 2 and 3 encode the alpha1 and alpha2 domains, which both bind the peptide, exon 4 encodes the alpha3 domain, exon 5 encodes the transmembrane region, and exons 6 and 7 encode the cytoplasmic tail. Polymorphisms within exon 2 and exon 3 are responsible for the peptide binding specificity of each class one molecule. Typing for these polymorphisms is routinely done for bone marrow and kidney transplantation. Hundreds of HLA-A alleles have been described. [provided by RefSeq, Jul 2008]
