Species Reactivity
Specificity
This assay has high sensitivity and excellent specificity for detection of HMGB1 . No significant cross-reactivity or interference between HMGB1 and analogues was observed. Note: Limited by current skills and knowledge, it is difficult for us to complete the crossreactivity detection between HMGB1 and all the analogues, therefore, cross reaction may still exist.

Intra-assay Precision
Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level HMGB1 were tested 20 times on one plate, respectively. Intra-Assay: CV<8%

Other Notes
Small volumes of HMGB1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS764105 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the High mobility group protein B1 (HMGB1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing HMGB1. The ELISA analytical biochemical technique of the MBS764105 kit is based on HMGB1 antibody-HMGB1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect HMGB1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, HMGB1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

HMGB1 elisa kit
Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- HMGB1 antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- HMGB1 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRPStreptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the HMGB1 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of HMGB1 can be calculated.

NCBI Summary for HMGB1
This gene encodes a protein that belongs to the High Mobility Group-box superfamily. The encoded non-histone, nuclear DNA-binding protein regulates transcription, and is involved in organization of DNA. This protein plays a role in several cellular processes, including inflammation, cell differentiation and tumor cell migration. Multiple pseudogenes of this gene have been identified. Alternative splicing results in multiple transcript variants that encode the same protein. [provided by RefSeq, Sep 2015]
UniProt Comments for HMGB1
HMGB1: DNA binding proteins that associates with chromatin and has the ability to bend DNA. Binds preferentially single-stranded DNA. Involved in V(D)J recombination by acting as a cofactor of the RAG complex. Acts by stimulating cleavage and RAG protein binding at the 23 bp spacer of conserved recombination signal sequences (RSS). Heparin-binding protein that has a role in the extension of neurite-type cytoplasmic processes in developing cells. Component of the RAG complex composed of core components RAG1 and RAG2, and associated component HMGB1 or HMGB2. Belongs to the HMGB family.
