MBS2025514 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the 11-Beta-Hydroxysteroid Dehydrogenase Type 1 (HSD11b1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing HSD11b1. The ELISA analytical biochemical technique of the MBS2025514 kit is based on HSD11b1 antibody-HSD11b1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect HSD11b1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, HSD11b1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to 11-Beta-Hydroxysteroid Dehydrogenase Type 1 (HSD11b1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to 11-Beta-Hydroxysteroid Dehydrogenase Type 1 (HSD11b1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain 11-Beta-Hydroxysteroid Dehydrogenase Type 1 (HSD11b1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of 11-Beta-Hydroxysteroid Dehydrogenase Type 1 (HSD11b1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene is a microsomal enzyme that catalyzes the conversion of the stress hormone cortisol to the inactive metabolite cortisone. In addition, the encoded protein can catalyze the reverse reaction, the conversion of cortisone to cortisol. Too much cortisol can lead to central obesity, and a particular variation in this gene has been associated with obesity and insulin resistance in children. Mutations in this gene and H6PD (hexose-6-phosphate dehydrogenase (glucose 1-dehydrogenase)) are the cause of cortisone reductase deficiency. Alternate splicing results in multiple transcript variants encoding the same protein.[provided by RefSeq, May 2011]
