The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing HSP-72. The ELISA analytical biochemical technique of the MBS705243 kit is based on HSP-72 antibody-HSP-72 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect HSP-72 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, HSP-72. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the direct competitive inhibition enzyme immunoassay technique. An antibody specific for HSP-72 has been pre-coated onto a microplate. Add HSP-72 (Standards or samples) to the well, and then add Biotin-conjugated HSP-72. A competitive inhibition reaction is launched between HSP-72 (Standards or samples) and Biotin conjugated HSP-72 with the HSP-72 antibody. Then add the HRP-avidin to each well. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of HSP-72 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This intronless gene encodes a 70kDa heat shock protein which is a member of the heat shock protein 70 family. In conjuction with other heat shock proteins, this protein stabilizes existing proteins against aggregation and mediates the folding of newly translated proteins in the cytosol and in organelles. It is also involved in the ubiquitin-proteasome pathway through interaction with the AU-rich element RNA-binding protein 1. The gene is located in the major histocompatibility complex class III region, in a cluster with two closely related genes which encode similar proteins. [provided by RefSeq, Jul 2008]
