The ELISA analytical biochemical technique of the MBS736390 kit is based on HSPA1A antibody-HSPA1A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect HSPA1A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, HSPA1A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the assay: HSP70k1A ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for HSP70k1A. Standards or samples are then added to the microtiter plate wells and HSP70k1A if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of HSP70k1A present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for HSP70k1A are added to each well to "sandwich" the HSP70k1A immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain HSP70k1A and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HSP70k1A concentration in each sample is interpolated from this standard curve.

This intronless gene encodes a 70kDa heat shock protein which is a member of the heat shock protein 70 family. In conjuction with other heat shock proteins, this protein stabilizes existing proteins against aggregation and mediates the folding of newly translated proteins in the cytosol and in organelles. It is also involved in the ubiquitin-proteasome pathway through interaction with the AU-rich element RNA-binding protein 1. The gene is located in the major histocompatibility complex class III region, in a cluster with two closely related genes which encode similar proteins. [provided by RefSeq, Jul 2008]
