This assay has high sensitivity and excellent specificity for detection of IGF-1R. No significant cross-reactivity or interference between IGF-1R and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between IGF-1R and all the analogues, therefore, cross reaction may still exist in some cases.

The ELISA analytical biochemical technique of the MBS7236768 kit is based on IGF1R antibody-IGF1R antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect IGF1R antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, IGF1R. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the Assay||IGF-1R ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-IGF-1R antibody and an IGF-1R-HRP conjugate. The assay sample and buffer are incubated together with IGF-1R-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IGF-1R concentration since IGF-1R from samples and IGF-1R-HRP conjugate compete for the anti-IGF-1R antibody binding site. Since the number of sites is limited, as more sites are occupied by IGF-1R from the sample, fewer sites are left to bind IGF-1R-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IGF-1R concentration in each sample is interpolated from this standard curve.
