The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing INB. The ELISA analytical biochemical technique of the MBS739085 kit is based on INB antibody-INB antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect INB antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, INB. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Intended Uses: This INHB ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey INHB. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: INHB ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-INHB antibody and an INHB-HRP conjugate. The assay sample and buffer are incubated together with INHB-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the INHB concentration since INHB from samples and INHB-HRP conjugate compete for the anti-INHB antibody binding site. Since the number of sites is limited, as more sites are occupied by INHB from the sample, fewer sites are left to bind INHB-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The INHB concentration in each sample is interpolated from this standard curve.

The inhibin beta B subunit joins the alpha subunit to form a pituitary FSH secretion inhibitor. Inhibin has been shown to regulate gonadal stromal cell proliferation negatively and to have tumour-suppressor activity. In addition, serum levels of inhibin have been shown to reflect the size of granulosa-cell tumors and can therefore be used as a marker for primary as well as recurrent disease. Because expression in gonadal and various extragonadal tissues may vary severalfold in a tissue-specific fashion, it is proposed that inhibin may be both a growth/differentiation factor and a hormone. Furthermore, the beta B subunit forms a homodimer, activin B, and also joins with the beta A subunit to form a heterodimer, activin AB, both of which stimulate FSH secretion.
