The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ISG15. The ELISA analytical biochemical technique of the MBS283284 kit is based on ISG15 antibody-ISG15 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ISG15 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ISG15. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate ISG15 in samples. An antibody specific for ISG15 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyISG15 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ISG15 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ISG15 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a ubiquitin-like protein that is conjugated to intracellular target proteins upon activation by interferon-alpha and interferon-beta. Several functions have been ascribed to the encoded protein, including chemotactic activity towards neutrophils, direction of ligated target proteins to intermediate filaments, cell-to-cell signaling, and antiviral activity during viral infections. While conjugates of this protein have been found to be noncovalently attached to intermediate filaments, this protein is sometimes secreted. [provided by RefSeq, Dec 2012]

Its enzymatic pathway is partially distinct from that of ubiquitin, differing in substrate specificity and interaction with ligating enzymes. ISG15 conjugation pathway uses a dedicated E1 enzyme, but seems to converge with the Ub conjugation pathway at the level of a specific E2 enzyme. Targets include STAT1, SERPINA3G/SPI2A, JAK1, MAPK3/ERK1, PLCG1, EIF2AK2/PKR, MX1/MxA, and RIG-1. Deconjugated by USP18/UBP43. Seems to display antiviral activity during viral infections. Interacts with, and is conjugated to its targets by the UBE1L (E1 enzyme) and UBE2E2 (E2 enzyme) (Probable).
