Intended Uses: This KCNQ2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human KCNQ2. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: KCNQ2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-KCNQ2 antibody and an KCNQ2-HRP conjugate. The assay sample and buffer are incubated together with KCNQ2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the KCNQ2 concentration since KCNQ2 from samples and KCNQ2-HRP conjugate compete for the anti-KCNQ2 antibody binding site. Since the number of sites is limited, as more sites are occupied by KCNQ2 from the sample, fewer sites are left to bind KCNQ2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The KCNQ2 concentration in each sample is interpolated from this standard curve.

Kv11.1: the ether-a-go-go related gene is a pore-forming (alpha) subunit of voltage-gated inwardly rectifying potassium channel is associated with cardiac arrhythmias and rhythmic excitability of the pituitary. Channel properties are modulated by cAMP and subunit assembly. Mediates the rapidly activating component of the delayed rectifying potassium current in heart (IKr). The potassium channel is probably composed of a homo- or heterotetrameric complex of pore-forming alpha subunits that can associate with modulating beta subunits. Heteromultimer with Kv11.2 and Kv11.3. Interacts with ALG10B. Heteromultimer with KCNE1 and KCNE2. Defects in Kv11.1 are the cause of long QT syndrome type 2 (LQT2), a heart disorder characterized by a prolonged QT interval on the ECG and polymorphic ventricular arrhythmias. They cause syncope and sudden death in response to exercise or emotional stress. Deafness is often associated with LQT2.
