The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing KLK14. The ELISA analytical biochemical technique of the MBS2023340 kit is based on KLK14 antibody-KLK14 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect KLK14 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, KLK14. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to Kallikrein 14 (KLK14) has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled Kallikrein 14 (KLK14) and unlabeled Kallikrein 14 (KLK14) (Standards or samples) with the pre-coated antibody specific to Kallikrein 14 (KLK14). After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of Kallikrein 14 (KLK14) in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of Kallikrein 14 (KLK14) in the sample.

This gene encodes a member of the kallikrein subfamily of serine proteases that have diverse physiological functions such as regulation of blood pressure and desquamation. The altered expression of this gene is implicated in the progression of different cancers including breast and prostate tumors. The encoded protein is a precursor that is proteolytically processed to generate the functional enzyme. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2015]
