We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of KYNU elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS2600302 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Kynureninase (KYNU) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing KYNU. The ELISA analytical biochemical technique of the MBS2600302 kit is based on KYNU antibody-KYNU antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect KYNU antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, KYNU. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This experiment use double-sandwich elisa technique and the ELISA Kit provided is typical. The pre-coated antibody is human KYNU monoclonal antibody and the detecting antibody is polyclonal antibody with biotin labeled. Samples and biotin labeling antibody are added into ELISA plate wells and washed out with PBS or TBS. Then Avidin-peroxidase conjugates are added to ELISA wells in order; Use TMB substrate for coloring after reactant thoroughly washed out by PBS or TBS. TMB turns into blue in peroxidase catalytic and finally turns into yellow under the action of acid. The color depth and the testing factors in samples are positively correlated.

UniProt Comments for KYNU
KYNU: Catalyzes the cleavage of L-kynurenine (L-Kyn) and L-3- hydroxykynurenine (L-3OHKyn) into anthranilic acid (AA) and 3- hydroxyanthranilic acid (3-OHAA), respectively. Has a preference for the L-3-hydroxy form. Also has cysteine-conjugate-beta-lyase activity. Xanthurenic aciduria manifesting as massive urinary excretion of large amounts of kynurenine, 3-hydroxykynurenine and xanthurenic acid has been observed in an individual carrying a homozygous missense change in KYNU (PubMed:17334708). The urinary pattern in the patient suggests kynureninase deficiency and a block in the conversion of kynurenine and 3-hydroxykynurenine to anthranilate and 3-hydroxyanthranilate, respectively. Belongs to the kynureninase family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Hydrolase; Mitochondrial; Amino Acid Metabolism - tryptophan; EC 3.7.1.3

Cellular Component: nucleoplasm; mitochondrion; cytoplasm; cytosol

Molecular Function: protein homodimerization activity; kynureninase activity; pyridoxal phosphate binding

Biological Process: anthranilate metabolic process; tryptophan catabolic process to acetyl-CoA; tryptophan catabolic process to kynurenine; quinolinate biosynthetic process; tryptophan catabolic process
