MBS2023241 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Leukocyte Associated Immunoglobulin Like Receptor 1 (LAIR1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing LAIR1. The ELISA analytical biochemical technique of the MBS2023241 kit is based on LAIR1 antibody-LAIR1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect LAIR1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, LAIR1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Leukocyte Associated Immunoglobulin Like Receptor 1 (LAIR1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Leukocyte Associated Immunoglobulin Like Receptor 1 (LAIR1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Leukocyte Associated Immunoglobulin Like Receptor 1 (LAIR1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Leukocyte Associated Immunoglobulin Like Receptor 1 (LAIR1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Inhibitory receptors regulate the immune response to prevent lysis of cells recognized as self. The gene is a member of both the immunoglobulin superfamily and the leukocyte-associated inhibitory receptor family. The gene maps to a region of 19q13.4 called the leukocyte receptor cluster, which contains at least 29 genes encoding leukocyte-expressed receptors of the immunoglobulin superfamily. The encoded protein has been identified as an anchor for tyrosine phosphatase SHP-1, and may induce cell death in myeloid leukemias. Alternative splicing results in multiple transcript variants.
