When a <span id=\"urn:enhancement-ebcc0533-e10d-4277-b553-03a2a0ca3344\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dna-sequence\">DNA sequence<\/span> is the foundation or code for a protein molecule, the particular <span id=\"urn:enhancement-422531ec-a0b1-4940-9da8-db1030121b62\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dna-strand\">DNA<\/span> molecule of interest can be blotted using Southern Blotting technique. It is done by detection of particular <span id=\"urn:enhancement-5d7b2be2-1e2f-478b-b584-f6e0cc791955\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> (or isolated mRNA).\u00a0 mRNA is generally represented as 5% of the overall <span id=\"urn:enhancement-b3fdfb1d-b12e-435b-830e-7ed100989419\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> sequence. This method reveals the identity, number, activity, and size of the particular <span id=\"urn:enhancement-00a40815-d6b3-4685-9a42-3fd0c04bd701\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/gene\">gene<\/span>. This blotting technique can also be used for the growth of a tissue or organism. In different stages of <span id=\"urn:enhancement-5efb8f47-5dac-422f-ba72-c6ed0f3a3cc4\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/differentiation\">differentiation<\/span> and morphogenesis the abundance of an <span id=\"urn:enhancement-46f3af63-4f65-4a7e-b3f6-c5741f709165\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> changes and this can be identified using this technique. It also aids in the identification of abnormal, diseased or infected condition at the molecular level. The northern blot technique was developed in 1977 by James Alwine, David Kemp and George Stank at Stanford University. The technique got its name due to the similarity of the process with Southern blotting. The primary difference between these two techniques is that northern blotting concerns only about <span id=\"urn:enhancement-f275085e-b074-41b6-8d44-808be8fefc3e\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span>.<\/p>\n<h3 style=\"text-align: left;\"><strong>Principle<\/strong><\/h3>\n<p style=\"text-align: left;\">As all normal blotting technique, northern blotting starts with the electrophoresis to separate <span id=\"urn:enhancement-39ad2b47-5c32-435f-b0bc-156a97550a7c\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> samples by size. Electrophoresis separates the <span id=\"urn:enhancement-828dac89-2ba2-4ccc-9686-8c465c348f10\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> molecules based on the charge of the nucleic acids. The charge in the nucleic acids is proportional to the size of the nucleic acid sequence. Thus the electrophoresis membrane separates the <span id=\"urn:enhancement-c430775b-bca3-46c3-9aeb-faae18b02f4f\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/nucleic-acid\">Nucleic acid<\/span> sequence according to the size of the <span id=\"urn:enhancement-cb7d3285-d35b-4685-a8be-1dff810db210\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> sequence. In cases where our target sequence is an mRNA, the sample can be isolated through oligo cellulose chromatographic techniques, as mRNA are characterized by the poly(A)-tail. Since gel molecules are fragile in nature, the separated sequences are transferred to the nylon membranes. The selection of nylon membrane is contributed to the factor that nucleic acids are negatively charged in nature. Once the <span id=\"urn:enhancement-6d4632b7-e167-4144-a3dc-46744677d320\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> molecules are transferred it is immobilized by covalent linkage. The probe is then added, the probe can be complementary an ss <span id=\"urn:enhancement-ae5d3350-0978-4ef8-ac17-b052ecf866aa\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/dna-sequence\">DNA sequence<\/span>. Formamide is generally used as a blotting buffer as it reduces the <span id=\"urn:enhancement-183cc09e-38e5-493d-88fc-ca6b89ed5401\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/tm\">annealing<\/span> temperature.<\/p>\n<h3 style=\"text-align: left;\"><strong>Procedure<\/strong><\/h3>\n<ol style=\"text-align: left;\">\n<li>The tissue or culture sample collected is first homogenized. The samples may be representative of different types of culture for comparison or it can be for the study of different stages of growth inside the culture.<\/li>\n<li>The <span id=\"urn:enhancement-80493520-63a3-4d66-a010-4e76c29238fc\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> sequence is separated in the electrophoresis unit an <span id=\"urn:enhancement-2708de59-454f-4792-b20b-b81b8c34962f\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/agarose\">agarose<\/span> gel is used for the purpose of the nucleic acid separation.<\/li>\n<li>Now the separated <span id=\"urn:enhancement-962aaacb-872f-4a7a-be1f-ebd3fb7e9c5c\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> sequence is transferred to the nylon membrane. This is done by two mechanisms capillary action and the ionic interaction.<\/li>\n<li>The transfer operation is done by keeping the gel in the following order. First, the <span id=\"urn:enhancement-70cbcc67-d774-4ff4-8ba4-c17e368f5106\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/agarose\">agarose<\/span> gel is placed on the bottom of the stack, followed by the blotting membrane. On top of these paper towels a mild weight (glass plate) is placed. The entire setup is kept in a beaker containing transfer buffer.<\/li>\n<li><span id=\"urn:enhancement-8c7169d5-f77d-43d6-b63c-0656f1e8d82a\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> transferred to the nylon membrane is then fixed using UV radiation.<\/li>\n<li>The fixed nylon membrane is then mixed with probes. The probes are specifically designed for the <span id=\"urn:enhancement-b3adebbd-09ee-4578-800d-639ae32301c7\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/gene\">gene<\/span> of interest, so that they will hybridize with <span id=\"urn:enhancement-9d9dabea-0967-48be-9492-c06e7107edac\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> sequences on the blot corresponding to the sequence of interest.<\/li>\n<li>The blot membrane is washed to remove unwanted probe<\/li>\n<li>Labeled probe is detected by chemiluminescence or autoradiography. The result will be dark bands in x ray film.<\/li>\n<\/ol>\n<h3 style=\"text-align: left;\"><strong>GEl and Probes<\/strong><\/h3>\n<p style=\"text-align: left;\">The <span id=\"urn:enhancement-c9e58d6a-59b5-4db2-9538-4020c90f2c77\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> samples are separated using <span id=\"urn:enhancement-7f915b51-a511-4ce3-8bf1-5a7272ce6d98\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/agarose\">agarose<\/span> gels using formaldehyde as <span id=\"urn:enhancement-f6c5dd37-dc9d-4433-8419-2440017f4e5c\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/denature\">denaturing<\/span> agents but in small <span id=\"urn:enhancement-90d0e9e0-a6b6-4b7f-b978-16fec9703311\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> or micro <span id=\"urn:enhancement-0d3481e9-20bf-41e1-b894-0b2ced3416d3\" class=\"textannotation disambiguated wl-creative-work\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/rna\">RNA<\/span> sequences, polyacrylamide sequences with urea as a <span id=\"urn:enhancement-8b427179-144e-4dbd-a706-7e9bf4debeb9\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/denature\">denaturing<\/span> agent also can be used. Ethidium bromide can be used as a staining agent. Two types of <span id=\"urn:enhancement-9a797ec2-8f8c-4586-9502-1c475b6eea59\" class=\"textannotation disambiguated wl-thing\" itemid=\"https:\/\/data.wordlift.io\/wl1503301\/entity\/markers\">markers<\/span> are for size marking.
